Spotted guppy

I purchased a few feeder guppies for my turtle in November. She decided she needed tank mates instead of a full belly. About 4 weeks ago, the largest female had babies. 4 of the 9 are still with me. About a week ago one lone baby appeared. I figure it was my smallest females first litter, since there was only one. 6 days ago, I drained, cleaned, and refilled the tank. Moving the guppies into a holding tank for the cleaning.The next day, I noticed a spot on my largest females tail. Her behavior was normal, and there is no other visible abnormalities. The day after, I woke to about 25 new babies. I still have about 18. Concerned that the spot could be ich, I started the pea method. It's been about 4 days now. The spot has not changed. All the fish seem to be thriving, and the fry are growing nicely. The female is not fully mature yet. Could this just be her color changing, or should I be concerned with her spot?
